What will you do?Title: Isolation of antigen-specific B cells from human samples and characterization of native antibodies produced by the selected cells.
Context: The candidate will be operative in the Preclinical research center, in the lab of B Cell Immunology & Ab Discovery. The team is dedicated to the analysis of the immune response to vaccination and infection, mainly oriented to the B cell immune response dissection and the BCR repertoire analysis. Moreover the team is involved in different projects aimed at the isolation of monoclonal antibodies and the development of new binding and functional assays for monoclonal antibodies and policlonal sera screening.
Project description: the project will be focused on the set up of new flow cytometric staining panels to identify different Ag-specific B cell subsets from which monoclonal antibodies can be isolated. Moreover new Luminex assays will be designed to evaluate the specificity of the antibodies released by the cells isolated and miniaturized functional assays will be set up to screen monoclonal and policlonal sera.
The activities related to these objectives are mainly summarized here:
Design of different panel of markers to identify multiple B cell subsets from human PMBC, mouse and rat lymphatic organs Training on the Luminex 3D technology and the miniatured multiplex assay Acquisition of analytical methods to screen for Ag-specificity culture surnatants by miniaturized Luminex assays Acquisition of technical ability to perform functional assays with specific pathogens
Competencies:
During the internship, the intern will develop:
Ability to analyze B cell markers expression by flow cytometry technology Acquisition of expertise in performing flow cytometric analysis Ability to prepare conjugated beads and setting the conditions for binding assaysby Luminex Assess Ag binding by Luminex assays Perform functional assay with at least one pathogen
